Artemether Can Attenuate Thrombus Formation by Inhibiting U46619-Induced Platelet Activation

Abstract
When blood vessels are damaged, a prompt hemostatic response follows to lower blood loss and maintain normal circulation, and platelet activation and aggregation are essential components of this response. Nevertheless, abnormal or excessive platelet aggregation underlies the etiologies of cardiovascular diseases such as thrombosis, atherosclerosis, and stroke. Therefore, the identification of substances that regulate platelet activation and suppress platelet aggregation is an important aspect of developments aimed at treating and preventing cardiovascular diseases. Artemether is a methyl ether derivative of artemisinin derived from the antimalarial plant Artemisia genus. We studied the effects of artemether on platelet functions using U46619-induced human platelets and found that artemether upregulated the cyclic adenosine monophosphate-dependent pathway and downregulated intracellular calcium mobilization, glycoprotein IIb/IIIa (¥áIIb/¥â3)-mediated signaling, and clot retraction. We suggest that artemether has effective antiplatelet and antithrombotic effects and is a potential therapeutic candidate for the prevention of platelet-induced cardiovascular disease.
